Antioxidant, Antimicrobial, and Tyrosinase Inhibitor Activities in the Aerial Parts of Onobrychis oxyodonta

Objective: The genus Onobrychis occurs widely across the world and occupies an important place among medical aromatic plants due to its antioxidant and phenolic contents. The aim of this study was to evaluate the constituents of phenolic acids, volatile components, antioxidant, antimicrobial, and tyrosinase inhibitor activities of Onobrychis oxyodonta.

Materials and Methods: 2,2-diphenyl-1-picrylhydrazyl free radical (DPPH∙), ferric reducing antioxidant power (FRAP), and total phenol content (TPC) were used to evaluate the antioxidant capabilities of the methanolic extract. The phenolic composition was investigated by means of reverse phase-high performance liquid chromatography (RP-HPLC). Antimicrobial activity was determined using the disc diffusion method. Tyrosinase inhibitor activity was tested using the colorimetric method.

Results: The IC50 value for DPPH assay was 1.333±0.0026 (mg/mL), the FRAP value 323±5.773 (μM Trolox/g sample) and the total phenolic content value 8.0±0.115 mg gallic acid per gram of sample. Ferulic acid, benzoic acid, sinapic acid, syringaldehyde were the main phenolic compounds in the extract. The half-maximal inhibitory concentrations (IC50) of the extract for tyrosinase were higher than for kojic acid. The extract exhibited moderate antibacterial activity against acid-fast bacterium (Mycobacterium smegmatis) and gram negative (Yersinia pseudotuberculosis) bacteria.

Conclusion: O. oxyodonta extract may be a useful pharmaceutical agent due to its powerful antioxidant, antimicrobial and moderate tyrosinase inhibitor activities.
