The Effect of DHEAS on Leptin, Lipid Profile, and Endothelial Function in the Rats Exposured To High-Fat Diet

According to previous reports, DHEAS has anti-atherosclerotic, and anti-obesity effects via regulating lipid metabolism, and decreasing body fat mass. This study aims to examine the effects of DHEAS on leptin, lipid profile, and endothelial function on the rats fed with high-fat diet (HFD).

Female Sprague-Dawley rats were divided into two groups: Group I (standard chow), -II (HFD). Following 5 months of feeding, group II were allocated into 3 subgroups: Group IIa (SF), -IIb (1 mg/kg DHEAS), and -IIc (10 mg/kg DHEAS). All treatments applied as (ip) for 7 days. ET-1, NO, and VEGF were measured in liver and aorta. Furthermore, TG, TC, HDL-C, and DHEAS were detected in serum.

No significant increase was observed in the body weights among the groups. Serum, liver, and aort leptin levels were found higher in HFD groups, although DHEAS did not lead to a difference. TG and TC increased in HFD groups. TG, TC, and HDL-C decreased with DHEAS. However, the decrease in HDL-C was insignificant. There was no effect of HFD on ET-1, NO, and VEGF. Significant increases was observed in serum ET-1, and NO with high-dose DHEAS. A significant decrease was noted only in ET-1 in liver. No difference was documented in the VEGF of serum, liver, and aorta. The mechanisms underlying effects of HFD and DHEAS on endothelial functions might be complex. As a consequence, it can be stated that DHEAS has an advantageous effect on lipid metabolism as it lowers TG and TC levels.
