The Effects of Vitamin E Derivatives on the Level of Glutathione in Experimentally Ischemia-Reperfusion Induced Guinea Pigs

The purpose of this study is to determine protective effects of vitamin E derivatives against oxygen radicals in guinea pig retina with ischemic injury. The right eyes of 40 male guinea pigs weighed 500-600 g were used and we divided them into 5 groups with randomly selected 8 guinea pigs. The animals were randomly assigned as group 1 (control), group 2 (I/R), group 3 (I/R + alpha- tocopherol), group 4 (I/R + gamma-tocopherol) and group 5 (I/R + d-alpha-tocopherol polyethylene glycol 1000 succinate, TPGS). Groups 2, 3, 4 and 5 were applied ischemia-reperfusion (I/R) and recevied serum physiologic, alpha-tocopherol, gamma-tocopherol and TPGS respectively. Retinal ischemia was induced for 90 minutes then followed by reperfusion for 24 hours. The animals were sacrificed at 24 hours of reperfusion. Then, retinas were isolated and processed for the quantification of glutathione (GSH) levels.

According to retinal GSH levels gamma-tocopherol did not have significant protective effect (p=0.53). Alpha-tocopherol and TPGS groups had significant protective effect but in the comparision of these two groups no statistically significant difference was determined (p=0.60).

Our results suggest that according to GSH levels in eyes that were induced to ischemia-reperfusion, vitamin E derivatives especially alpha-tocopherol and TPGS can have protective role by decreasing lipid peroxidation.
