Determination of iNOS and PARP Immunoreactivity in Rats Kidney Tissues With Experimentally Induced Diabetes

Objective: Diabetic nephropathy is an important complication of diabetes mellitus. Nitric oxide (NO) is an effective multifunctional mediator for hemodynamic variance in diabetes. PoliADP ribose polymerase (PARP) is an enzyme that has a role in DNA repairing during apoptotic processes. Excessive activation of PARP results in apoptosis. In this study, it was purposed to examine the inducible nitric oxide synthase (iNOS) and PARP immunoreactivities in kidney tissues of experimental diabetic rats induced by Streptozotosin.

Materials and Methods: Total of 14 male Wistar Albino rats were used in this study. Rats were divided in to the two groups as group I (Control) and Group II (Diabetes). In the Diabetes group, a single dose of 60 mg/kg STZ, which is dissolved in phosphate-citrate buffer (pH: 4.5) was injected intraperitoneally. All rats in both groups were decapitated under anesthesia and kidney tissues were removed and fixed in 10% formaldehyde solution for light microscopic investigations at the end of the 10 weeks. Paraffin blocks were prepared and 5 μm tissue sections were cut and stained with avidin-biotin peroxidase method to analyse iNOS and PARP immunoreactivities. The severity and prevalance of immunoreactivities were evaluated semiquantitatively by scoring from 0 to +3.

Results: In the light microscopic examination, diabetic grup II showed prominent +3 iNOS and PARP immunoreactivity whereas immunoreactivities of control group I were less and +1.

Conclusion: Thus, we concluded that iNOS and PARP, influence on apoptotic process, have important contributions on kidney damage in the diabetic group.
