Experimental Evaluation of the Effects of Losartan on Osseointegration of Titanium Implants

Objective: Losartan has been reported to reduce osteoclastogenesis and increase osteoblast activity. The aim of this study was to evaluate the effect of systemic losartan application on titanium implant osseointegration.

Materials and Methods: Titanium implants were surgically integrated in the tibial bones of rats, after which the rats were randomly divided into two groups: a control group (n=10) and a losartan treatment group (n=10). No other treatment was applied to the controls, whereas losartan was administered to the treatment group in a dosage of 30 mg/kg by oral gavage with saline solution three times per week during an eight-week experimental period. After the experimental period, titanium implants were collected with the surrounding bone tissues and fixed. Non-decalcified histological analyses of implants and surrounding bone tissue were performed. The bone implant connection ratio (%) of each implant was calculated as the ratio of the length of the implant circumference to the circumference of the total implant in direct contact with the bone. The bone-filling ratio (%) was determined by the ratio of bone-filled areas within the region to the total area of the region, including the area 0.5 mm from the titanium implant in periimplant bone.

Results: Although the bone implant connection and bone-filling ratios (%) ratios were higher in the losartan group than in the controls, there was no statistically significant difference in the ratios between the two groups (P>0.05).

Conclusion: Within the limitations of this study, losartan increased the osseointegration levels numerically but not statistically.
