Investigation of the Effect of Paracetamol on Spontaneous Bladder Detrusor Smooth Muscle Contractions in Female Rats

Objective: Although paracetamol, a pharmacological agent with a wide spectrum of use due to its analgesic and antipyretic effects, is frequently used in diseases affecting the urinary system, its effect on bladder contraction was unknown. This study aimed to reveal the effect of paracetamol on the contraction of the detrusor smooth muscle.

Materials and Methods: Bladder strips (2 mm x 10 mm) obtained from intact female Wistar-albino rats in the diestrus phase were suspended in an isolated organ bath containing 5 mL of Krebs solution at 37 ⁰C and pH 7.4 under a tension of 1.5 grams (n=7). Following the regulation of spontaneous contractions, paracetamol at doses of 1000 μM and 2000 μM was added in a non-cumulative manner. The peak-to-peak values and the area under the curve values obtained before and after the application were standardized as percentage changes. Data were analyzed using Paired t-test in SPSS 22.0 program.

Results: It was found that the application of paracetamol significantly inhibited the spontaneous contraction of the detrusor smooth muscle at doses of 1000 μM and 2000 μM (p<0.05).

Conclusion: Paracetamol administration has a dose-dependent inhibitory effect on the contraction of the detrusor smooth muscle, and considering this effect is clinically important by using paracetamol in individuals with urinary system pathologies and various other indications.
