In this study, production and characterization of monoclonal antibody (Mabs) to heamagglutinin protein of rinderpest virus (RPV) were aimed. In present study, initially, splenocytes from the mice immunized with RPV-RBOK and myeloma cell line (FO) were fused by polyethylene glycol-dimethyl sulfoxide (PEG/DMSO). For the selection of hybrid cell lines, the cell culture media containing hypoxanthine-aminopterin-thymidine (HAT) was used. An enzyme linked immunosorbent assay (ELISA) was utilized to select hybrid cell line producing antibodies against RPV. After limiting dilution procedure, sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and immunblotting were used to determine the specifity of anti-H Mab. These antibodies were characterized for certain properties such as sub-type and neutralization affect. With this study, production and characterization of monoclonal antibody (Mabs) to heamagglutinin protein of rinderpest virus (RPV) was carried out.
