Effects of Lycopene on Cisplatin and Gentamicin Induced Oxidative Stres in Blood and Liver in Rats

This study was carried out to establish the effects of lycopene on administrations of high dose cisplatin and gentamicin induced oxidative stres in blood and liver and to investigate whether lycopene have any ameloriate effects.

Rats were divided into 5 groups, each consisting of 6 rats. Group 1 used as control. Animals of group 2, 3, 4 and 5 were administrated single dose cisplatin (7 mg/kg); cisplatin following 5 days lycopene (4 mg/kg); gentamicin for 6 days (100 mg/kg) and gentamicin together with lycopene respectively. Blood and liver tissue samples were collected 5 days after single dose cisplatin and 24 hours after the last treatment in other groups. The levels of malondialdehyde and reduced glutathione levels were determined spectrophotometrically.

Significant increases in plasma MDA, blood GSH and liver MDA, levels were observed in single dose cisplatin treated rats. Administration of lycopene after cisplatin produced decreases in these parameters. No change was observed liver GSH levels in these groups compared to the control group. While significantly increase was determined alone the levels of plasma MDA 6 days treatment of gentamicin in group 4 when compared control group; liver MDA, GSH and blood GSH levels were not changed. Normalization of plasma MDA levels was determined in treated lycopene together with gentamicin group.

In conclusion, treatment with gentamicine were not affected on oxidative stres in blood and liver, but cisplatin increased in rats, and it was concluded that lycopene may prevent increasing oxidative stres in blood and liver.
