Presence of –SH Groups in Liver Tissue Arginase of Sheep and Effects of Amino Acids Containing Sulphur Group, Aromatic and Hydroxylic Amino Acids on the Enzyme Activity

The effects on sheep liver tissue arginase of tyrosine and tryptophan from aromatic amino acids; serine and threonine from hydroxylic amino acids; cysteine and methionine from amino acids containing sulphur group and homocysteine from amino acids that do not occur in proteins were investigated. To understand whether there was thiol groups in enzyme structure or not, effects of p-chloromercuri benzoate and n-ethylmaleimide was also investigated.

As a result of the investigation, while arginase was inhibited by cysteine, homocysteine and thyroptophan, was less activated by serine and threonine; methionine and tyrosine did not cause any changes on arginase activity. While p-chloromercuri benzoate, cysteine and homocysteine caused activation in low concentration, they caused inhibition in high concentration.

Cysteine, homocysteine and thyriptophan from amino acids caused mixed inhibition, p-chloromercuri benzoate caused uncompetetive inhibition and n-ethylmaleimide caused noncompetetive inhibition.
