In this study, it was aimed to make a comparative examination of rumen content samples taken from sheep with different methods in terms of pH and to investigate the differences between the results.

For this purpose, 20 male sheep of Akkaraman breed which were clinically healthy, aged 1–1.5, living weight of 35–45 kg and brought to slaughterhouse for slaughter were used.

Rumen content samples were taken from all animals that were pre-slaughtered after clinical examinations by using the rumen probe used in our clinic, vacuum rumen probe and rumen puncture (rumenosynthesis), and the sample was taken from the same animals directly after slaughter. Rumen content samples taken from each animal in 4 ways, pH was determined separately with pH meter and it was evaluated whether there were significant differences between the methods.

The average pH values of rumen probe, vacuum probe, rumenosynthesis and rumen content taken after slaughter were 6.48±0.36, 6.33±0.38, 6.23±0.37 and 6.15±0.30, respectively. The pH values obtained by the rumen probe were significantly higher than those obtained by other methods (P<0.05); the difference between the vacuum tube and those taken with rumeocentesis was not significant; the difference between the vacuum tube and the postslaughter values was significant; It was determined that there was no significant difference between pH values of rumenosentez and those taken after slaughter.

As a result; it has been concluded that the rumen catheter can be used in routine ruminant clinics in order to obtain rumen content, but it is more appropriate to use vacuum rumen catheter in scientific studies.
